Mechanism of Action Voriconazole is a broad-spectrum antifungal agent that inhibits the synthesis of ergosterol, a key component of fungal cell membranes, thereby disrupting fungal cell integrity and growth.
Indications Voriconazole is used to treat serious fungal infections, including invasive aspergillosis, candidemia, esophageal candidiasis, and other severe fungal infections caused by susceptible species.
Dosage Forms Available as tablets (50 mg, 200 mg), oral suspension (40 mg/mL), and injection.
Storage Conditions Tablets and oral suspension should be stored at room temperature (68-77°F or 20-25°C) and protected from light. The injection form is usually administered in a hospital or clinic.
Common Side Effects Rash, headache, nausea, vomiting, diarrhea, visual disturbances (e.g., blurred vision, color changes), photosensitivity, and liver function abnormalities.
Serious Side Effects Severe skin reactions (e.g., Stevens-Johnson Syndrome, Toxic Epidermal Necrolysis), kidney damage, adrenal gland problems, bone pain, and pancreatitis.
Drug Interactions Voriconazole interacts with many medications, including corticosteroids, QT-prolonging drugs, sirolimus, rifampin, carbamazepine, phenytoin, ritonavir, and St. John’s wort.
Special Precautions Avoid direct sunlight and use sunscreen due to photosensitivity. Regular liver function monitoring is recommended. Patients should avoid alcohol and high-fat foods.
Pregnancy and Lactation Voriconazole can harm the unborn baby and is not recommended during pregnancy or breastfeeding
