Mechanism of Action TIC-10 is a dual inhibitor of Akt and ERK, leading to the upregulation of the tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) through Foxo3a. It induces TRAIL-mediated apoptosis in cancer cells.
Biological Activity – In Vitro: Causes a dose-dependent increase in TRAIL mRNA and protein localization on the cell surface of cancer cell lines. It induces cell death in TRAIL-sensitive cancer cells but spares normal fibroblasts.
- In Vivo: Induces tumor regression in various cancer xenograft models, including colon cancer and triple-negative breast cancer.
Applications – Research: Studied for its effects on cancer cell apoptosis and tumor regression.
- Medicine: Investigated as a potential antitumor therapeutic agent for various cancers, including glioblastoma multiforme.
Physical Properties – Appearance: White to beige solid.
- Solubility: Soluble in DMSO (2 mg/mL, clear upon heating).
- Storage: 2-8°C.
Safety – Handle with care to avoid contact with skin and eyes.
- Store in a cool, dry place.
