Pharmacokinetics	– Absorption: Rapidly absorbed after oral administration, with peak plasma concentrations reached within 3 to 4 hours.<br>- Distribution: Volume of distribution is approximately 1.1 L/kg.<br>- Metabolism: Metabolized in the liver via CYP2C19 and CYP3A4.<br>- Elimination: Half-life is approximately 1 hour (shorter in fast metabolizers, longer in slow metabolizers).<br>- Excretion: Primarily excreted in urine.
Side Effects	– Common: Headache, nausea, vomiting, diarrhea, constipation, abdominal pain, flatulence, dizziness.<br>- Serious: Acute tubulointerstitial nephritis (kidney inflammation), Clostridioides difficile infection (severe diarrhea), severe skin reactions (e.g., Stevens-Johnson syndrome, toxic epidermal necrolysis), hypomagnesemia (low magnesium levels), vitamin B12 deficiency, increased risk of bone fractures.<br>- Allergic Reactions: Anaphylaxis, rash, swelling, difficulty breathing.
Precautions	– Monitor for signs of kidney problems, severe diarrhea, or skin reactions.<br>- Regular blood tests may be needed to check for vitamin B12 deficiency or hypomagnesemia.<br>- Use with caution in patients with liver disease.<br>- Avoid concomitant use with rilpivirine-containing products.<br>- Inform healthcare providers of all medications and supplements being taken.
