Description
Pyrazinamide is a white or almost white, crystalline powder. It is sparingly soluble in water, slightly soluble in ethanol (96 per cent) and in methylene chloride. It shows polymorphism.
Application
Pyrazinamide is an anti-tuberculosis drug. It has been used as a second-line drug in the past and is often used in retreatment patients with other anti-tuberculosis drugs. Used in combination with other anti-tuberculosis drugs to treat tuberculosis that is ineffective by first-line anti-tuberculosis drugs (such as streptomycin, isoniazid, rifampicin, and ethambutol).

|
Melting point is approximately 189 – 191 °C. Sparingly soluble in water, soluble in ethanol, and slightly soluble in ether. It remains stable under normal conditions but may decompose under extreme temperature or in contact with strong acids/bases.

Pharmacology
|
An essential anti – tuberculosis drug. It works by inhibiting the growth of Mycobacterium tuberculosis, especially effective against intracellular bacteria. It penetrates the bacterial cell membrane and disrupts their metabolism. Usually used in combination with other anti – TB drugs like isoniazid, rifampicin, and ethambutol to enhance treatment efficacy and prevent drug resistance.

|
Commonly causes liver function impairment, indicated by elevated transaminase levels. It may also lead to hyperuricemia, which can result in joint pain and potentially gout. Gastrointestinal issues such as nausea, vomiting, and loss of appetite are also possible. Rarely, allergic reactions and hematological problems may occur.

Dosage depends on factors like patient’s age, weight, and condition. Typically, adults take 15 – 30 mg/kg daily, divided into 3 – 4 oral doses. The treatment course usually lasts several months as part of a combination therapy.
