Mechanism of Action	Pirfenidone exhibits both anti-inflammatory and antifibrotic effects. It inhibits the production and activity of transforming growth factor-β (TGF-β), a key factor in fibrosis. It also suppresses the expression of inflammatory cytokines such as TNF-α, IL-4, and IL-13. Additionally, it upregulates matrix metalloproteinases (MMPs) to reduce extracellular matrix (ECM) accumulation.
Clinical Uses	Pirfenidone is primarily used to slow the progression of idiopathic pulmonary fibrosis (IPF) and is one of the two conditionally recommended drugs for this condition. It is also being evaluated for other fibrosing lung diseases and potential use in cardiac disorders with fibrosis.
Pharmacokinetics	Pirfenidone is rapidly absorbed in the gastrointestinal tract with a half-life of about 2–2.5 hours. It is metabolized in the liver, primarily through the cytochrome P450 system, and excreted mainly as the metabolite 5-carboxy-pirfenidone.
Side Effects	Common side effects include nausea, vomiting, loss of appetite, dizziness, and photosensitivity.
