Mechanism of Action	Hydroxyurea is a ribonucleotide reductase inhibitor that selectively targets the S phase of the cell cycle, inhibiting DNA synthesis and cell proliferation.
Indications	– Treatment of chronic myeloid leukemia.<br>- Treatment of ovarian cancer.<br>- Treatment of certain types of skin cancer (squamous cell cancer of the head and neck).<br>- Reduction of painful crises and blood transfusion needs in sickle cell anemia.<br>- Other off-label uses include treatment of melanoma, renal cancer, and psoriasis.
Dosage Forms	Oral capsule (200 mg, 300 mg, 400 mg, 500 mg), oral tablet (100 mg, 1000 mg).
Administration	– Take orally with or without food, usually once daily.<br>- Swallow capsules whole; do not crush, chew, or open.<br>- Tablets can be swallowed with water or dissolved in a small amount of water.<br>- Liquid form should be measured carefully with a special device.
Side Effects	– Serious Side Effects: Numbness or tingling in hands or feet, skin numbness or purple discoloration, skin ulcers, sudden chest pain, low blood cell counts, signs of liver or pancreas problems.<br>- Common Side Effects: Loss of appetite, nausea, constipation, diarrhea, low blood cell counts, bleeding, mouth sores.<br>- Other Side Effects: Bone marrow suppression (leading to anemia, leukopenia, and thrombocytopenia), gastrointestinal reactions (nausea, vomiting, constipation), skin and vascular toxicity (ulcers, necrosis), teratogenic effects, alopecia, skin pigmentation, weight gain.
Contraindications	– Hypersensitivity to hydroxyurea.<br>- Pregnancy (due to potential teratogenic effects).<br>- Concurrent use with live vaccines.
Special Considerations	– Both men and women should use contraception during treatment and for a specified period afterward to prevent pregnancy.<br>- Increased risk of developing leukemia or skin cancer with long-term use.<br>- Monitor blood counts regularly to detect bone marrow suppression.<br>- Avoid contact with the drug if pregnant or planning to become pregnant.<br>- Use with caution in patients with kidney or liver disease.<br>- May interact with antiviral drugs and interferon.
