Mechanism of Action	Fluticasone propionate is a potent corticosteroid that exerts its effects by binding to glucocorticoid receptors. Pharmacokinetics	– Absorption: Systemic bioavailability is low after oral inhalation (<1%) and intranasal use (<2%). Most of the inhaled dose is swallowed and undergoes extensive first-pass metabolism.<br>- Distribution: Highly protein-bound (99%) in serum. Volume of distribution is 4.2 L/kg.<br>- Metabolism: Extensively metabolized in the liver by cytochrome P450 3A4 (CYP3A4), forming inactive metabolites.<br>- Excretion: Primarily eliminated via feces, with <5% excreted in urine.<br>- Half-Life: Terminal half-life is approximately 7.8 to 10.8 hours.
Safety Profile	– Systemic Effects: Low risk of systemic side effects such as hypothalamic-pituitary-adrenal (HPA) axis suppression due to low systemic absorption.<br>- Local Effects: May cause local side effects such as oral thrush or skin atrophy with prolonged topical use.<br>- Special Populations: Caution advised in patients with liver impairment due to the drug’s extensive hepatic metabolism.
