Econazole nitrate is a pyrrole antifungal drug and a dechlorinated derivative of miconazole. It has antibacterial activity against Candida, chromomycetes, coccidiospores, histoplasm, Sporothrix, and Trichophyton.
Description
Econazole nitrate is a white or almost white, crystalline powder. It is very slightly soluble in water, soluble in methanol, sparingly soluble in methylene chloride, slightly soluble in ethanol (96 per cent).
Application
Econazole nitrate (CAS 68797-31-9) can be used for the treatment of cutaneous candidiasis. It can also be used to treat body lice, hand lice, femurs, athlete’s foot, and tinea versicolor.

Melting point: Around 160 – 162 °C. Solubility: Slightly soluble in water, freely soluble in dimethylformamide, soluble in methanol, and sparingly soluble in ethanol. It is stable under normal storage conditions but may decompose when exposed to high humidity, extreme heat, or strong light over an extended period.

Pharmacology
|
Econazole nitrate is an antifungal agent. It works by inhibiting the synthesis of ergosterol, an essential component of the fungal cell membrane. By interfering with ergosterol production, it disrupts the integrity and function of the fungal cell membrane, leading to the death of the fungus. It is effective against a wide range of dermatophytes, yeasts, and some molds. It is commonly used in the treatment of superficial fungal infections of the skin and mucous membranes, such as athlete’s foot, ringworm, and vaginal candidiasis.

Side Effects
|
Topical application may cause mild local skin reactions in some individuals, including itching, burning, redness, and rash. These side effects are usually transient and resolve on their own. In rare cases, more severe allergic reactions may occur. When used intravaginally, it can sometimes cause vaginal irritation or a change in vaginal discharge. Systemic absorption is generally low, but in cases of overuse or in patients with compromised skin barriers, there is a potential for more systemic side effects, although these are extremely rare.

Usage
|
For topical treatment of skin infections, it is available as creams, ointments, and solutions. The affected area is typically cleaned and dried before applying a thin layer of the econazole nitrate – containing product, usually 1 – 2 times a day for a specified duration, which can range from a few weeks to several months depending on the type and severity of the infection. For vaginal candidiasis, it is available as vaginal suppositories or creams, and the dosage and treatment duration are clearly specified in the product instructions, often involving nightly applications for a week.

Store in a well – closed container, protected from light, and in a cool place (preferably between 15 – 30 °C). Avoid storing in areas with high humidity to maintain its stability and potency.
