Mechanism of Action	Deflazacort is a corticosteroid prodrug that is rapidly converted to its active metabolite, 21-deflazacort. It binds to glucocorticoid receptors, exerting anti-inflammatory and immunosuppressive effects.
Pharmacokinetics	– Absorption: Rapidly absorbed after oral administration, with peak plasma concentration occurring within 1-2 hours.<br>- Distribution: Volume of distribution is approximately 204 ± 84 L.<br>- Metabolism: Deacetylated by plasma esterases to 21-deflazacort, further metabolized by CYP3A4.<br>- Elimination: Primarily excreted in urine (70%) and feces (30%) within 24 hours.<br>- Half-Life: 1.1 to 1.9 hours.
Indications	Used to treat Duchenne muscular dystrophy (DMD) in patients aged 5 years and older. It is also used off-label for conditions such as rheumatoid arthritis, nephrotic syndrome, systemic lupus erythematosus, and transplantation.
Adverse Effects	Common side effects include increased risk of infections, hypertension, fluid retention, behavioral and mood disturbances, decreased bone mineral density, and ophthalmic effects such as cataracts.
Storage Conditions	Store at room temperature (20°C to 25°C; 68°F to 77°F) with excursions permitted between 15°C to 30°C (59°F to 86°F). Discard any unused oral suspension after 1 month of first opening the bottle.
Special Considerations	– Gradual Dose Reduction: Abrupt discontinuation may lead to adrenal insufficiency.<br>- Infection Risk: Patients should avoid exposure to chickenpox or measles and seek medical advice if exposed.<br>- Cardiovascular/Renal Effects: Monitor blood pressure and consider dietary salt restriction if fluid retention occurs.<br>- Bone Health: Prolonged use may increase the risk of osteoporosis.<br>- Growth in Children: Deflazacort may have less negative impact on growth compared to other corticosteroids.
