Mechanism of Action	Amiloride hydrochloride dihydrate inhibits the epithelial sodium channels (ENaC) in the distal nephron, lung, and colon. It selectively blocks sodium transport, reducing potassium, hydrogen, calcium, and magnesium secretion. It also has potential vasodilatory effects.
Pharmacokinetics	Absorption: Onset of action within 2 hours, peak plasma concentrations achieved within 3–4 hours.<br> Distribution: High volume of distribution (350–380 L) due to low plasma protein binding (<40%).<br> Metabolism: Not metabolized by the liver; excreted unchanged by the kidneys.<br> Elimination: 50% excreted in urine, 40% in feces; serum half-life of 6–9 hours.
Therapeutic Uses	Used as a potassium-sparing diuretic for hypertension and edematous states. It can also be used in combination with thiazides or loop diuretics to reduce the risk of hypokalemia.
