Definition Rifaximin is a non-systemic, orally administered antibiotic that is poorly absorbed and primarily acts within the gastrointestinal tract.
Mechanism of Action – It inhibits bacterial RNA synthesis by binding to the beta-subunit of bacterial DNA-dependent RNA polymerase, preventing transcription.<br>- It has broad-spectrum activity against both Gram-positive and Gram-negative bacteria.
Clinical Uses – FDA-Approved Indications:<br> – Treatment of traveler’s diarrhea caused by noninvasive strains of Escherichia coli in patients ≥12 years.<br> – Reduction of overt hepatic encephalopathy recurrence in adults.<br> – Treatment of irritable bowel syndrome with diarrhea (IBS-D) in adults.<br>- Off-Label Uses:<br> – Small intestinal bacterial overgrowth (SIBO).<br> – Diverticulitis.<br> – Inflammatory bowel disease and pouchitis.<br> – Prophylaxis of spontaneous bacterial peritonitis.
Dosage Forms – Tablets: 200 mg, 550 mg.
Storage Conditions Store at room temperature (68°F to 77°F or 20°C to 25°C), away from heat, moisture, and direct light.
