Name & CAS
Menadione sodium bisulfite (CAS 130 – 37 – 0). Its chemical name is 2 – Methyl – 1,4 – naphthoquinone sodium bisulfite. The molecular formula is \(C_{11}H_9NaO_5S\), and it has a molecular weight of 296.24 g/mol. It appears as a yellow – orange crystalline powder.
Properties
Solubility: Freely soluble in water, slightly soluble in ethanol. It is hygroscopic, meaning it readily absorbs moisture from the air. In solid form, it is relatively stable under normal conditions but may degrade when exposed to heat, strong acids, or alkalis. The solution may also decompose over time, especially when exposed to light.

Pharmacology
Menadione sodium bisulfite is a synthetic form of vitamin K. Vitamin K plays a crucial role in the blood – clotting cascade by helping in the synthesis of several clotting factors, such as prothrombin. It is used to treat vitamin K deficiency, which can lead to bleeding disorders. In some cases, it may be prescribed to patients with liver diseases or those on certain medications that interfere with vitamin K metabolism. However, excessive intake of menadione sodium bisulfite can have adverse effects as it can be converted to a more toxic form in the body.
Side Effects
High doses of menadione sodium bisulfite can cause hemolytic anemia, especially in individuals with glucose – 6 – phosphate dehydrogenase (G6PD) deficiency. It can also lead to jaundice in newborns if given inappropriately. Gastrointestinal discomfort, including nausea and vomiting, may occur in some patients. There are concerns about its potential carcinogenicity, although more research is needed to fully understand this risk.
Usage
It is available in oral and injectable forms. For oral use, it is typically given in tablet or capsule form to correct vitamin K deficiency. In cases of severe deficiency or when oral administration is not possible, it can be administered intramuscularly or subcutaneously. The dosage is carefully determined based on the patient’s age, weight, and the severity of the vitamin K deficiency. For example, in adults with mild deficiency, the daily oral dose may range from 10 – 20 mg, while in more severe cases, higher doses or parenteral administration may be required.
Storage
Store in a tightly closed container, protected from light, and in a dry place. A cool environment (preferably below 25 °C) helps maintain its stability. Due to its hygroscopic nature, it should be kept away from areas with high humidity to prevent clumping and degradation.
