Famciclovir is an anti herpesvirus drug. It is the first oral drug approved in the United States for facial and genital herpes. It is also the only drug to reduce post herpetic neuralgia.

Description

Famciclovir is a white or off-white crystalline powder. It is mainly used for herpes virus infection, especially herpes zoster.

Application

It is used for herpes virus infectious diseases, such as chickenpox, herpes zoster, genital herpes and herpes simplex. It is particularly practical for acyclovir to treat viral infections that are not ideal.

Mechanism of Action Famciclovir is a prodrug that is rapidly converted to its active form, penciclovir, in the body. Penciclovir is phosphorylated by viral thymidine kinase in infected cells to form penciclovir triphosphate, which competitively inhibits viral DNA polymerase, leading to chain termination and inhibition of viral replication.
Indications – Treatment of acute herpes zoster (shingles) caused by varicella-zoster virus (VZV).<br>- Management of genital herpes simplex virus (HSV) infections, including initial and recurrent episodes.<br>- Suppressive therapy for chronic genital HSV infections.<br>- Potential use in treating chronic hepatitis B infections.
Pharmacokinetics – Absorption: Rapidly absorbed orally, with peak plasma concentrations of penciclovir reached within 1 hour.<br>- Distribution: Penciclovir has a volume of distribution of 1.08 L/kg.<br>- Metabolism: Converted to penciclovir via first-pass metabolism; metabolites are inactive.<br>- Excretion: Approximately 94% of the dose is excreted in urine within 24 hours.
Adverse Effects – Common: Headache, nausea, diarrhea, dyspepsia.<br>- Rare: Hepatotoxicity, Steven-Johnson syndrome.<br>- No significant toxic side effects reported, and no antidote is needed.
Contraindications – Hypersensitivity to famciclovir or penciclovir.<br>- Severe renal impairment (dose adjustment required).
Occupational Exposure Famciclovir has an occupational exposure limit (OEL) and acceptable daily exposure (ADE) value to ensure safety in manufacturing environments.
