| Mechanism of Action | Axitinib is a potent and highly selective inhibitor of vascular endothelial growth factor receptor (VEGFR) tyrosine kinases 1, 2, and 3. It competitively binds to the intracellular ATP site of VEGFRs, inhibiting signal transduction and reducing angiogenesis. |
| Indications | Axitinib is indicated for the treatment of advanced renal cell carcinoma (RCC) after failure of one prior systemic therapy. |
| Dosage Forms | Available as oral tablets in 1 mg and 5 mg strengths. |
| Recommended Dosing | The starting dose is 5 mg orally twice daily, approximately 12 hours apart, with or without food. |
| Common Side Effects | Hypertension, diarrhea, fatigue, decreased appetite, nausea, dysphonia, palmar-plantar erythrodysesthesia syndrome (hand-foot syndrome), weight loss, vomiting, asthenia, and constipation. |
| Serious Side Effects | Arterial and venous thromboembolic events, hemorrhage, gastrointestinal perforation and fistula formation, thyroid dysfunction, reversible posterior leukoencephalopathy syndrome (RPLS), and wound healing complications. |

| Special Populations | Use with caution in patients with hepatic impairment. Axitinib has not been studied in patients with severe hepatic impairment. |
